APOE and frontotemporal dementia Lead Investigator: Rahul Desikan Institution : UCSD E-Mail : rahuldesikan@gmail.com Proposal ID : 599 Proposal Description: A substantial amount of prior work indicates that the E2 and E4 alleles of apolipoprotein E (APOE) impact Alzheimer's disease (AD) age of onset. However, the relationship between APOE and frontotemporal dementia (FTD) has not been evaluated in a large, neuropathologically-confirmed, population based cohort. Additionally, the relationship between APOE and cognitive changes and the impact of cardiovascular risk factors on APOE-associated FTD risk factors is not known. In this proposal, using the large population based NACC cohort, we will evaluate the relationship between the E2, E3 and E4 APOE alleles and the various FTD subtypes. We will additionally evaluate the relationship between cognitive changes and APOE on FTD associated neuropathology as well as the role of cardiovascular risk factors, in combination with APOE alleles, on impacting FTD age of onset.
